1 M MOPS (pH 7.0) recipe
A Good's buffer for pH 6.5–7.9 (pKa 7.2), used in bacterial media, protein work and RNA gel buffers.
🧮 Volume calculator
| Component | Amount | Final concentration (1X) |
|---|---|---|
| MOPS (free acid) | 20.93 g | 1 M |
| 10 M NaOH | as needed | to pH 7.0 |
| Water (distilled) | to final volume |
Steps
- Dissolve in about 80% of the final volume of water.
- Adjust to pH 7.0 with NaOH.
- Bring to the final volume with water.
- Filter-sterilise through 0.22 µm (do not autoclave). Store at 4 °C in the dark.
Good to know
- MOPS yellows when autoclaved or exposed to light; pale yellow is fine, dark yellow should be discarded.
FAQ
Is this the MOPS buffer for RNA gels?
No — RNA gels use a 10X MOPS running buffer with sodium acetate and EDTA.
This is a widely used standard composition. Amounts depend on the hydrate form and purity of your reagents — check the formula weight (FW) on the bottle and follow your institution's safety rules and the manufacturers' instructions.
