YPD medium (yeast) recipe
The standard rich medium for yeast (S. cerevisiae): 1% yeast extract, 2% peptone, 2% glucose.

Steps
- Dissolve yeast extract and peptone in 90% of the final volume and autoclave.
- Sterilise glucose separately as a 20–40% stock and add after cooling.
- For plates add 20 g/L agar.
Good to know
- Autoclaving glucose with the rest browns the medium (Maillard reaction) and slows growth slightly.

YPAD adds adenine (typically 40 mg/L) to prevent red pigment and poor growth in ade2 strains.
