SDS-PAGE running buffer (Tris-glycine-SDS) recipe
Fills the SDS-PAGE tank. 1X is 25 mM Tris, 192 mM glycine and 0.1% SDS (pH ≈ 8.3, not adjusted).

Concentrated stocks (e.g. 10X) contain every component at that multiple — dilute to 1X for use.
Steps
- Dissolve Tris and glycine in about 80% of the final volume.
- Add SDS and mix gently to avoid foam.
- Bring to volume. Do not adjust the pH.
Good to know
- Adjusting the pH with HCl changes the ion composition and distorts bands.
- If SDS precipitates in the 10X stock, warm it before diluting.

Can I reuse running buffer?
Some labs reuse the outer chamber once or twice but always refill the inner chamber. Replace it if bands smile or run slowly.
