dNTP mix (10 mM each) recipe
A PCR dNTP mix made from 100 mM dATP, dCTP, dGTP and dTTP stocks. PCR usually uses 0.2 mM each (1 µL per 50 µL reaction).

| Component | Amount | Final concentration (1X) |
|---|---|---|
| 100 mM dATP | 100 µL | 10 mM |
| 100 mM dCTP | 100 µL | 10 mM |
| 100 mM dGTP | 100 µL | 10 mM |
| 100 mM dTTP | 100 µL | 10 mM |
| Nuclease-free water | to final volume |
Steps
- On ice, combine the four thawed dNTPs with water.
- Aliquot 50–100 µL and store at −20 °C.
Good to know
- Repeated freeze–thaw degrades dNTPs (keep to ≤10 cycles).
- dNTPs chelate Mg²⁺ — if you raise dNTPs substantially, raise MgCl₂ too.
FAQ
Is "10 mM total" different from "10 mM each"?
Yes. 10 mM each = 40 mM total; 10 mM total = 2.5 mM each. Always check the label.
This is a widely used standard composition. Amounts depend on the hydrate form and purity of your reagents — check the formula weight (FW) on the bottle and follow your institution's safety rules and the manufacturers' instructions.
