RNase A 10 mg/mL (DNase-free stock) recipe
An RNase A stock for removing RNA from plasmid and genomic DNA preps; boiling destroys any contaminating DNase.
🧮 Volume calculator
| Component | Amount | Final concentration (1X) |
|---|---|---|
| RNase A (bovine pancreas) | 100 mg | 10 mg/mL |
| 10 mM Tris-HCl pH 7.5 + 15 mM NaCl | to final volume |
Steps
- Dissolve RNase A in 10 mM Tris-HCl pH 7.5, 15 mM NaCl.
- Boil for 15 min and cool slowly to room temperature (RNase A refolds; DNases do not).
- Aliquot and store at −20 °C.
Good to know
- RNase A is extremely stable — keep it away from RNA work areas and pipettes.
FAQ
Doesn't boiling destroy RNase too?
RNase A has many disulfide bonds and refolds into its active form on cooling.
This is a widely used standard composition. Amounts depend on the hydrate form and purity of your reagents — check the formula weight (FW) on the bottle and follow your institution's safety rules and the manufacturers' instructions.
